crRNA Design & Synthesis
crRNA consists of two structural components: one part forms the hairpin structure that binds to the Cas effector, while the other part is a "protospacer" sequence (typically 20 bases) that specifically recognizes the target gene fragment. The crRNA binds to the Cas protein and the target gene fragment to form a Cas-crRNA-target ternary complex, after which the target sequence is cleaved by the Cas protein. Depending on the detection system and the type of Cas protein, different crRNAs can be designed to meet the detection requirements. For instance, Cas12a is guided by crRNA to specifically recognize and cleave dsDNA targets with a PAM sequence (5' TTTN-3'), while Cas13a, guided by crRNA, recognizes and cleaves ssRNA targets without requiring PAMs for target recognition.
